Characterization of Microsomal Glutathione-Dependent Peroxidation Inhibiting Factor of the Rat liver

Abstract
This study was aimed to characterize further the glutathione-dependent peroxidation inhibiting factor (GPIF) which has recently been proposed by several investigators to act as a unique antioxidant in the rat liver microsomes. When microsomes were heat-denatured at 100¡É for 5 min, the GPIF activity disappeared completely, but the level of -tocopherol decreased only slightly. The GPIF activity was not detected in liposomes made of phospholipid and ¥á-tocopherol. When microsomes containing 0.1 mM GSH were oxidized by meta-catalyzed oxidation system, GSH was consumed completely first, and then ¥á-tocopherol began to be used. And lipid peroxidation began to occur after the ¥á-tocopherol level was decreased to about 70%. These results suggest that microsomal GPIF may be an antioxidant protein that recycles tocopheroxyl radical to ¥á-tocopherol in membranes, using GSH as a cosubstrate
